SARS-CoV-2 Detection: Fast and Cost-Effective Sample Processing Prior to RT-PCR

The pandemic COVID-19 needs a rapid microbiological diagnostic from Clinical Microbiology Units. Due to the fact that it is done by using reverse transcription polymerase chain reaction (rRT-PCR) previous RNA extraction and automated equipment reagents for represent an economic increase diagnosis, we describe easy, cost-effective fast alternative extraction-free SARS-CoV-2. Samples were treated with proteinase K 10 minutes at 55°C. Then, there heat-process 5 98°C finally, 3 -20°C before commercial-commonly-used rRT-PCR procedure. automated-extraction was also performed QIAsymphony Kit (Qiagen) equipment. A total of 220 nasopharyngeal oropharyngeal swabs analyzed. 113 samples tested positive whereas 106 negative method, agreement 99%. one discordant sample noted in which no amplified result (gene ORF1ab N) observed gene (Ct 39) N 37) extraction-free. Thus, results comparable automated-extraction.
